HERVolution Data: HERV-K Expressed Across 43 Cancer Types

COPENHAGEN – October 29, 2026 -- HERVolution Therapeutics ApS published peer-reviewed data in the Journal of Translational Medicine showing HERV-K expression across the majority of 43 cancer types in RNA sequencing data from 1,353 patients, backing the target rationale for its lead program, IPT-001.

HERV-K expression spans nearly all cancer types examined

The analysis used data from the Copenhagen Prospective Personalized Oncology study (CoPPO, NCT02290522), which enrolled patients with advanced, predominantly pre-treated cancer. Most patients showed more than 100 HML-2 reads per million total reads across the 43 cancer types, cancers of unknown primary and unclassified tumors. The authors say this extends earlier indication-specific reports.

Single Q525A substitution turns a tolerated self-antigen into an immunogen

A systematic mutational screen of the immunosuppressive domain of the HERV-K envelope protein identified Q525A, termed ISDmut, as the most potent activator of NF-κB signaling. The authors describe it as the first immunostimulatory ISD mutation reported for a β-retrovirus. In human monocyte-derived dendritic cells, ISDmut increased envelope antigen expression and shifted the transcriptional program from cellular senescence toward antigen presentation and programmed cell death pathways. Virus-like particle assembly and prefusion envelope conformation were preserved, and the co-expressed GAG-ENV cassette abrogated envelope fusion activity, a safety-relevant parameter.

Human CD8+ T cells recognize HERV-K-expressing lung cancer cells

Dendritic cells transduced with adenoviral vectors encoding HERV-K-ISDmut expanded CD4+ and CD8+ T cells from healthy donors. In an HLA-matched setting, the expanded CD8+ T cells recognized the non-small cell lung cancer (NSCLC) cell line HCC827. HLA class I blockade partially reversed the response. HERVolution plans a first-in-human study in NSCLC.

Animal models show broken tolerance and improved survival

Adenoviral and mRNA-LNP vectors encoding HERV-K-ISDmut elicited strong T cell and antibody responses in mice and broke tolerance in HERV-K transgenic animals. In an exploratory cynomolgus macaque study, immunization raised HERV-K-specific antibodies that bound the envelope surface unit. In a murine metastatic renal carcinoma model expressing HERV-K, prime-boost immunization significantly improved lung metastasis clearance and overall survival versus control, with added benefit from combination with anti-PD-1 therapy.

FDA clearance of a third-party HERV-K vaccine IND sets regulatory precedent

In January 2026, the U.S. Food and Drug Administration cleared an Investigational New Drug application for a third-party self-amplifying RNA cancer vaccine encoding the HERV-K envelope protein. The first patient was dosed in July 2026 in a Phase 1 triple-negative breast cancer study. HERVolution differentiates through its engineered ISDmut antigen, designed to overcome immune tolerance to native HERV-K sequences, and through its reach across oncology, metabolic disease and cellular senescence.

CEO J. Robert Coleman said the data reinforce the company's view that the "dark genome" is a therapeutically actionable target class. He added that unpublished metabolic disease and senescence data are due soon.

The study was conducted with the University of Copenhagen, Rigshospitalet, Herlev Hospital, IPB University and Revvity Gene Delivery GmbH. Funding came from HERVolution, the Serum Institute of India, the European Innovation Council Accelerator, the EUREKA Eurostars Programme and Innovation Fund Denmark.
