Tel Aviv and Oslo – September 15, 2026 -- BioLineRx Ltd. (NASDAQ/TASE: BLRX) and Hemispherian AS will present two abstracts on their oncology candidate GLIX1 at the 21st Meeting of the European Association of Neuro-Oncology (EANO 2026) in Rome, Italy, September 24-27, with one abstract selected for oral presentation.
Preclinical data show GLIX1 penetrates the brain at 68-85% of plasma levels
The mini-oral presentation, scheduled for September 25, will detail preclinical efficacy of GLIX1, an oral TET2 activator, across two orthotopic glioblastoma xenograft models -- SNB19 and the more aggressive U87-MG -- as well as a temozolomide-resistant patient-derived xenograft model. Oral dosing ranging from 75 mg/kg once daily to 1000 mg/kg twice daily produced dose-dependent tumor growth inhibition and survival benefit at all tested doses, with temozolomide used as a positive control. Brain exposure reached 68-85% of plasma levels in mice, supporting central nervous system penetration critical for glioblastoma treatment.
Phase 1/2a trial is actively enrolling patients across four U.S. academic sites
A second abstract, presented as an e-poster, outlines the design of the ongoing first-in-human Phase 1/2a study (NCT07464925) evaluating GLIX1 in recurrent or progressive high-grade glioma. The open-label, multicenter trial uses a BOIN design to guide dose escalation across up to five dose levels in as many as 30 patients with WHO grade 3/4 glioma who have received no more than two prior therapy lines. Primary endpoints include safety, tolerability, and determination of the maximum tolerated dose or recommended Phase 2 dose; secondary endpoints cover pharmacokinetics, antitumor response, and six-month progression-free survival.
GLIX1 targets DNA damage response in a cancer type with reduced 5hmC levels
Glioblastoma tumors characteristically show markedly reduced genomic 5-hydroxymethylcytosine (5hmC) compared with healthy brain tissue, reflecting impaired TET2 activity. GLIX1 activates TET2 to drive DNA demethylation, generating single-stranded DNA breaks that mature into lethal double-stranded breaks, exploiting this vulnerability. Exploratory endpoints in the ongoing trial include pharmacodynamic markers P21, TMEM71, 5hmC, 5mC, and γH2AX.
Dr. Adam Robertson, Chief Scientific Officer of Hemispherian, will present the preclinical mini-oral session, while Dr. Ditte Primdahl of Northwestern Medicine will present the trial-design poster.
